A Lewis A trisaccharide (Galβ1-3[Fucα1-4]GlcNAc) equipped with an azide-functionalized linker through a short propyl spacer. The terminal azide can be covalently coupled to alkyne partners via copper-catalyzed azide-alkyne cycloaddition (CuAAC), or to dibenzocyclooctyne (DBCO) and related strained alkynes using copper-free click chemistry, forming stable 1,2,3-triazole linkages.
Lewis A is a histo-blood group antigen and a ligand for carbohydrate-binding proteins, including DC-SIGN and selected galectins. It serves as the biosynthetic precursor to sialyl-Lewis A (CA19-9), a clinically important selectin ligand and tumor-associated antigen, making Lewis A a useful probe for glycosylation and lectin-recognition studies. This probe can be used as a soluble inhibitor or immobilized on plates or sensors to investigate binding, and it is a substrate for sialyltransferases and fucosidases.
