Lewis X Propyl-Amine

A Lewis X trisaccharide (Galβ1-4[Fucα1-3]GlcNAc) equipped with an amine-functionalized linker through a short propyl spacer. The primary amine can be covalently coupled with N-hydroxysuccinimide (NHS) activated esters to form stable amide bonds, providing a straightforward route to conjugate the glycan onto NHS-activated molecules and surfaces. Lewis X is a histo-blood group-related antigen, also known as the stage-specific embryonic antigen SSEA-1 (CD15), and is a ligand for carbohydrate-binding proteins including DC-SIGN. It plays roles in cell adhesion, embryonic development, and immune recognition. It serves as the biosynthetic precursor to sialyl-Lewis X, a key selectin ligand, making Lewis X a useful probe for glycosylation and lectin-recognition studies. This probe can be used as a soluble inhibitor or immobilized on plates or sensors to investigate binding, and it is a substrate for sialyltransferases and fucosidases.
